Isolation and characterisation of porcine circovirus type-2 from sera of stillborn foetuses

PCV2 as a foetal pathogen is considered in this study. Sera from stillborn foetuses (collected on a farm with a history of reproductive failure) were examined for presence of PCV2 antibodies. Twenty-eight of the 171 sera tested were PCV2 antibody positive. Of these 28 positive sera, 13 were found to contain PCV2 viral DNA by PCR. Virus isolation was then performed on these samples positive for both PCV2 antibody and viral DNA. PCV2 was isolated in 9 sera. Lastly, amplification and sequencing of two isolates from stillborn foetuses showed 98% similarity. The homology of the two isolates with other PCV2 isolates was confirmed but comparison with the genomes of PCV2 isolates associated with PMWS indicated small differences. This study brings evidence of a foetal infection with PCV2 in relation with reproductive failure under field conditions. Further investigations are necessary to explain the differences in genomes.
