Association of lymphopenia with porcine circovirus type 2 induced postweaning multisystemic wasting syndrome (PMWS)

The precise identification of leucocyte subpopulations in the leukopenia induced by PCV2 infection and PMWS is the aim of the study. Ten piglets were experimentally infected with PCV2 and ten other were sham-inoculated. Peripheral blood samples were collected from day 1 post infection to day 35 and monitored. Four PCV2-infected piglets developed PMWS and were dead by day 25 post infection. The leukopenia developed by PMWS-affected piglets was mainly a lymphopenia that was notable from day 14 post infection. The characterization of this lymphopenia was made by single and triple labelling. While all B and T cell populations were reduced, the most discernible depletion was observed for CD21+B cells and memory/activated Th cells (CD3+, CD4+, CD8+). Also NK cells were depleted. Control animals and non-symptomatic PCV2-infected animals did not suffer from leukopenia.
