Virology

Porcine Reproductive and Respiratory Syndrome (PRRS)

DELPUTTE PL, NAUWYNCK HJ
Porcine arterivirus infection of alveolar macrophages is mediated by sialic acid on the virus.
Journal of Virology, 2004, Volume 78, Nº15, 8094-8101

In a previous study (VANDERHEIJDEN N et al., Journal of Virology, 2003, Vol. 77, Nº15, 8207-8215, see Bibliographical Bulletin of October 2003), the role of porcine sialoadhesin (pSn) in the internalization of PRRSV into porcine alveolar macrophages (PAMs) was clearly demonstrated. The objective of this follow-up study was to determine whether pSn has the capacity to bind sialic acids and to understand to what extent the presence of sialic acids on PRRSV is linked to the attachment to pSn and infection of PAMs. On the one hand, pSn was shown to be a lectin and to bind sialic acids. On the other hand, the removal of sialic acids from PPRSV through enzymatic treatment prevented infection and reduced attachment to PAMs indicating that such mechanisms depend on the presence of sialic acids on the virus.
