Swine Influenza

Protection of weaned pigs by vaccination with human adenovirus 5 recombinant viruses expressing the hemagglutinin and the nucleoprotein of H3N2 swine influenza virus.

The genes encoding for the hemagglutinin (HA) and nucleoprotein (NP) of the H3N2 subtype of swine influenza were inserted into a human adenovirus (Ad5) which was used as vector. The experiment included five groups of 3-week-old pigs as follows: three groups of 10 pigs were vaccinated intramuscularly with a recombinant adenovirus expressing the NP, with a recombinant adenovirus expressing the HA or with both recombinant viruses, while the two other groups were left unvaccinated. All groups except one control group were challenged with a virulent H3N2 field strain. Complete protection was achieved in the group vaccinated with both recombinant viruses (expressing HA and NP) as indicated by the lack of nasal shedding of virus and lung lesions following challenge. This finding is particularly interesting for very young piglets since this type of vaccination is not hampered by maternally-derived antibodies.
