Analysis of the subcellular localization of the proteins Rep, Rep' and Cap of porcine circovirus type 1.

The localization of the Cap (structural protein), Rep and Rep’ (replication proteins) of PCV1 was investigated using immune fluorescence techniques so as to better understand the processes of replication and maturation of the virus. While Rep and Rep’ were both found to reside only in the nucleus but never in the nucleoli, Cap was detected both in the nucleoplasm together with the replication proteins and in the nucleoli. Moreover, a redistribution of Cap from the nucleoli to the nucleoplasm was demonstrated. A potential sequence of the events is thus hypothesized for PCV1 infection: entry of Cap into the nucleoli, accumulation of Cap, Rep and Rep’ in the nucleoplasm, viral replication, capsid formation and encapsidation still in the nuceoplasm, followed by translocation to the cytoplasm and release of the virions.
