Development of a reverse transcription-PCR assay to detect porcine circovirus type 2 transcription as a measure of replication.

This assay was demonstrated to detect a nucleotide fragment corresponding to a capsid mRNA during PCV2 replication, allowing to distinguish between PCV1 and PCV2 on one hand and between RNA products and viral DNA of PCV2 on the other hand. This assay has several additional advantages: it is a single-tube protocol and hence avoids cross-contamination, it is specific, sensitive and much more rapid compared to other techniques such as virus titration, in situ hybridization or Southern blot.
