The ORF3 protein of porcine circovirus type 2 is involved in viral pathogenesis in vivo.

In a previous study carried out in vitro in PK15 cells, the open reading frame 3 (ORF3) protein of porcine circovirus type 2 (PCV2) had been shown to be involved in the apoptosis process induced by the virus without being necessary for its replication. To further investigate this feature of PCV2, an ORF3-deficient mutant PCV2 (mPCV2) was designed and inoculated to BALB/c mice. Several parameters were considered and compared between mPCV2 inoculated mice, wild-type PCV2 (wPCV2) inoculated mice and control mice: PCV2 antibody levels, PCV2 genomic copy numbers, microscopic lesions, amounts of viral DNA and antigen in lymph nodes, levels of CD4+ and CD8+ cells. The results of this study carried out in vivo confirm that PCV2 can replicate without ORF3 protein and that PCV2 pathogenesis greatly relies on ORF3 protein.
