Validation of serum samples pooling efficacy in regards of the diagnosis of PRRS virus by ELISA and QRT-PCR.
Proceedings of the 19th IPVS Congress, Copenhagen, Denmark, 2006 - Volume 2:33
The objective of this study was to determine the efficacy of serum samples pooling in the detection of porcine reproductive and respiratory syndrome virus (PRRSV). This technique is used to reduce the costs induced by regular monitoring of pig herds for diseases. The study was conducted on MARC-145 cells infected with a VR-2332-PRRSV strain and 5 serum sample poolings were tested in total. Analyses were carried out (TCID50 technique, qRT-PCR, ands ELISA tests) in order to get reliable data on this technique. The results indicated that this technique was reliable when PCR was used, as its sensitivity was not altered, though this technique might not be the best option in all contexts. Also, pooling serum samples and analyzing them with ELISA did not prove satisfactory, as a decrease was observed in the test sensitivity.
